In the manual it says that there is a high level ability called Assassination that makes the thief automatically backstab every few attacks. Does that work also with the swashbuckler kit, who normally can't backstab?

No. A Swashbuckler gets access to the warrior HLA Whirlwind instead, because he/she has no backstab multiplier.

He/ she also gets +hit, +damage and -armour which helps with the two weapon fighting style.
