Yes. Anyone can shoot at any time, and they can shoot at any living player. When a mod checks the thread, all shots that have been 'fired' will be checked in order of submission to see whether each shot is a miss (*click*) or a hit (*bang*). A hit means that the targeted player is dead and any further shots in the queue are discarded.
Once every living player has taken a shot (and missed), or once a shot has been fired (meaning someone is now dead), the queue will be reset and everyone is free to try shooting again.
You are free to discuss (or not) and try to fire the gun at any time once the game starts. There is no Night Phase, so the scum thread will be open for the entire game. The game ends once there is only one faction left alive (scum don't auto-win once they reach number parity with town).

The mod will show up at least periodically. All valid shots will be accepted even if a mod isn't present, and they'll be reviewed for success whenever a mod shows up.
Early in the game, when a lot of people are still alive, the queue will generally be reset by someone's death. Once there are only a few players left, the queue may be reset because every living player has taken a shot and missed.
