Your mana pool empties at the end of each phase in a turn.
Mana sources and instants may be used at any time, but other actions such as casting sorceries, summoning creatures, and attacking may only be performed during your turn.
Phases in brief:
1) Beginning Phase (untap, upkeep, draw)
2) First Main Phase (land, sorceries, enchantments, artifacts, creatures)
3) Combat Phase (attack)
4) Second Main Phase (sorceries, enchantments, artifacts, creatures)
5) End Phase
Usually, the beginning and ending of these phases do not have to be explicitly declared. Players need to understand the order in which a turn proceeds, but actual play is rather smooth in general.
What all that means for you: You should use the Wall of Roots activated ability only when you need to generate mana to cast a spell (and as per the text of the card, you may only activate it once per turn), and only when you cannot get the mana from a land or other source (otherwise you should avoid weakening the Wall of Roots).
